When handling and processing seeds the user should be aware that many seeds contain very toxic properties and must be dealt with according to 'Good Laboratory Practice'. The grinding of seeds should be carried out in an extraction cabinet and a face mask and gloves should be worn during this process. Fluid extracts may be poisonous and every care must be exercised when processing these fluids.
Inverclyde hold stocks of seeds suitable for lectin extraction. Where possible all lectin material will be shown to have the appropriate activity before it is delivered to a customer. The following are generally available.
Dolichos biflorus (Horsegram) - This seed yields an excellent anti-A1 reagent. It also contains anti-Tn and anti-Cad.
Ulex europaeus (Common Gorse) - A useful source of anti-H suitable for cell typing and secretor status work.
Bauhinia alba, Bauhinea Purpurea, Bauhinea variegata - This group of seeds contain anti-N and anti-Tn.
Laburnum alpinum - This seed contains anti-H.
Sophora japonica (Pagoda tree) - Extracts contain anti-A and anti-B. It reacts strongly with En(a-) red cells and can differentiate human from animal blood stains.
Ricinus communis (castor bean) - Contains a host of agglutinins and was the first know lectin.
Bandierea simplicifolia - also known as
Griffonia opposofolia, extracts contain anti-B and sometimes a very weak anti-A. Can be split into 5 fractions and a useful anti-Tk and anti-HbM can be found in BSII. BSI contains anti-Tn.
Phaseolus lunatus - Contains anti-A1.
Glycine soja or max - Contains anti-T, -Tn and -Cad. It is a very worthwhile lectin when incorporated into a polyagglutination panel. It can also be used for the control of enzymes.
Salvia group - This group of seeds is widely used to differentiate and identify polyagglutinable red cells.
Arachis hypogea (Peanut) - Contains anti-T and anti-Tk. It is also reported to contain anti-Th and anti-Tx.
Leonuris cardiaca - Contains ant-Cad and a weak anti-T which can be diluted out.
Mollucella laevis - Contains anti-A and anti-N and has also been reported to contain anti-Tn.
Hibiscus sabdariffa (Jamaican sorrel) - Contains anti-T, anti-Tn and anti-Th.
Parthenocissus tricuspidata - This extract has been reported to contain anti-C3b.
Vicia unijuga - The leaves of this plant contain an excellent anti-N.
Fomes fomentarius - A freshly extracted reagent can provide a potent anti-B. Collected from birch trees in Northern Scotland.
