
Improper fixation of the red blood cells on a glass slide can interfere with elution of the red cells. This will result in some of the red cell taking up some of the counterstain and appearing pink or pale red rather than giving the expected ghost like appearance. While the experienced eye is unlikely to confuse these cells with the strongly stained fetal red cells, less experienced staff might be exposed to a problem. This difficulty is usually caused by an alcohol fixative which has become de-stabilised.
FIXOL is a stabilised buffered fixative reagent which will overcome the above difficulties. It has a 2 year expiry from the date of manufacture and has the advantage that the drying time can be cut to 2 minutes and the fixing time to 3 minutes.
